Cormlet Formation in Gladiolus Shoot Base by Growth Retardants in vitro


Abstract
This experiment was carried out to investigate the effective cormlet formation condition in vitro. The result of experiment for inducing direct cormlet formation from shoot-base of in vitro plantlets of gladiolus (cv. Topaz) was that the percentage of cormlet formation was the highest by 65.8% in primary liquid medium with 10 ¥ìM CCC. In the secondary solidified cormlet-induction medium containing 0.7 - 1.1% agar, 6-9% sucrose, 1-10 ¥ìM GA©ý, and 10 ¥ìM NAA and 250 ¥ìM BA, the formation of cormlet was highly increased. The fresh weight per cormlet was remarkably increased by adding 10 ¥ìM CCC, 0.5% agar, 3-9% sucrose, 10 ¥ìM GA©ý, missing ABA, and NAA and BA. The percentages of cormlet formation and fresh weight of the solidified cormlet-induction medium without ABA were 76.2% and 60.5 §·, respectively. this medium was most effective for improving formation and growth of cormlets of all the treatments. Days required for cormlet formation was about 28 days in all the media, the number of cormlets per shoot were not different among treatments. The sprouting percentage of cormlet formed in vitro was the highest by 68.4% and the days required for sprouting of cormlets was shorter by 21.1 days in room temperatures than in temperature treatment at 4¡É.
